Small-molecule IGF1R inhibitor healthspan program
preclinicalEvaluate whether orally delivered small-molecule IGF1R inhibitors can improve healthspan and potentially lifespan as an anti-aging therapy.
C57BL/6 mouse longevity experiment using dietary administration of selective IGF1R inhibitors picropodophyllin (PPP) or NVP-ADW742 starting at 13 months of age, with physiological, behavioral, frailty, and survival assessments.
Preclinical mouse healthspan study reported in a 2026 bioRxiv preprint.
Both compounds protected against short-term memory decline and improved several physiological or frailty-related measures. PPP was associated with gastrointestinal bleeding toxicity. NVP-ADW742 produced a squarer survival curve indicating 93-day longer healthspan, but drug-likeness analysis suggested potential cardiotoxicity and brain bioaccumulation.
