Antibodies sustain platelet activation
Full text
Could estradiol, an estrogen hormone, leave a clotting risk when treatment resumes despite avoiding the liver? This hypothesis asks whether plasma antibodies, immune proteins, carry it.
The test would expose donor platelets, clot-forming blood fragments, to post-resumption plasma at fixed estradiol and platelet factor 4 (PF4), a platelet protein. It predicts excess platelet activation, a step in clot formation.
Removing immunoglobulin, another name for antibodies, should erase activation; adding back patient immunoglobulin should restore it. Blocking FcγRIIa, a platelet receptor activated by antibody complexes, or removing PF4-binding antibodies should also end it. This transfer-loss-rescue pattern would favour the hypothesis over antibody-independent explanations.
