This proposed study asks why cellular senescence, a lasting halt in cell division, might return after senescent cells are removed. Its hypothesis centres on LINE-1, a mobile genetic element that can copy itself into DNA. The proposed sequence is that LINE-1 activity and DNA damage rise in formerly dividing cells, then lead to persistent arrest and a secretory state. The decisive test would suppress LINE-1, then restore ORF2, its DNA-cutting protein. If active ORF2 restarts recurrent senescence while an inactive form at comparable expression leaves recurrence suppressed, that pattern would support this proposed intracellular route. Testing requires tracking DNA breaks, new LINE-1 insertions, cell fate and signals between tissues over time.
